# GLP-1 pills compared: Rybelsus, Wegovy tablets and Foundayo (orforglipron) in 2026

By [the GLPMate team](/editorial-policy) · Based on FDA prescribing information & peer-reviewed sources · Updated 2026-10-14 · 7 min read

Quick answer

The US has three kinds of GLP-1 pill in 2026: oral semaglutide for type 2 diabetes (Rybelsus, Ozempic tablets), Wegovy tablets for weight and Foundayo (orforglipron) for weight. Semaglutide pills need an empty stomach and a 30-minute wait; Foundayo has no food or water rules. In separate trials, weight fell 13.6% (Wegovy tablets) and 11.1% (Foundayo top dose). Your prescriber decides which fits.

## What GLP-1 pills are available in 2026?

For years, the only GLP-1 pill was oral semaglutide for type 2 diabetes. That changed fast. In the US in 2026 there are three kinds of GLP-1 pill:

- **Oral semaglutide for type 2 diabetes:** Rybelsus (3, 7 and 14 mg) and, since early 2026, Ozempic tablets (1.5, 4 and 9 mg), a newer formulation on the same label.
- **Wegovy tablets:** oral semaglutide 25 mg for weight management, approved in December 2025.
- **Foundayo (orforglipron):** a small-molecule GLP-1 pill from Lilly for weight management, approved on 1 April 2026.

We checked each current US label on DailyMed on 14 October 2026. Labels change, so your prescriber and pharmacist have the final word. If you already take one of these pills, our guide to [tracking a daily oral GLP-1](/blog/oral-glp-1-tracking-2026) covers the daily routine in more depth.

## How do Rybelsus, Wegovy tablets and Foundayo compare?

Pill | US use | Dose steps (once daily) | How to take | Pivotal trial result

**Rybelsus** (oral semaglutide) | Type 2 diabetes; lowers heart-event risk in high-risk adults | 3 mg for 30 days, then 7 mg; 14 mg if more glucose control is needed | Empty stomach, up to 4 oz water, wait 30 min | 26 weeks: HbA1c −1.2 (7 mg) and −1.4 points (14 mg) vs −0.3 with placebo

**Ozempic tablets** (oral semaglutide) | Same as Rybelsus | 1.5 mg for 30 days, then 4 mg; 9 mg if needed | Same as Rybelsus | Same label as Rybelsus; doses are not interchangeable mg for mg

**Wegovy tablets** (oral semaglutide) | Weight management; heart-risk reduction in adults with heart disease and overweight | 1.5, 4 and 9 mg (30 days each), then 25 mg | Same as Rybelsus | OASIS 4, 64 weeks: −13.6% body weight vs −2.4% with placebo

**Foundayo** (orforglipron) | Weight management | 0.8, 2.5, then 5.5 mg; then 9, 14.5 or 17.2 mg, at least 30 days per step | With or without food, any time of day | ATTAIN-1, 72 weeks: −11.1% at 17.2 mg vs −2.1% with placebo

The trial results come from different studies with different people and lengths. They are not a head-to-head comparison, and the table shouldn’t be read as a ranking.

## How do you take oral semaglutide vs Foundayo?

This is the biggest day-to-day difference between the pills.

### Oral semaglutide: strict morning routine

All three semaglutide pills (Rybelsus, Ozempic tablets and Wegovy tablets) share the same rules in their labels:

- Take one tablet in the morning on an empty stomach, with up to 4 ounces (about 120 mL) of plain water and no other drink.
- Swallow it whole. Don’t split, crush or chew it.
- Wait at least 30 minutes before eating, drinking anything else or taking other oral medicines.
- If you miss a dose, skip it and take the next one the following day. Never take two in one day.

The reason is absorption. The Wegovy label puts oral semaglutide’s bioavailability at only about 1–2%. In the label’s studies, absorption was higher with a small sip of water than with a full glass, and a longer wait before eating raised it further.

**30 minutes** The minimum wait after an oral semaglutide tablet (Rybelsus, Ozempic tablets, Wegovy tablets) before food, other drinks or other oral medicines, per the current US labels. Foundayo's label sets no such wait.

One more label point: in a pharmacology study, oral semaglutide raised levothyroxine exposure by about a third. If you take thyroid medication, tell your prescriber.

### Foundayo: no food or water rules

The Foundayo label says to take one tablet once daily, with or without food, at any time of day. It sets no fasting window or water limit. A few other rules matter instead:

- **Missed dose:** take it as soon as possible, but never two in one day. If you miss 7 or more days in a row, your prescriber may restart you at a lower dose.
- **Drug interactions:** strong CYP3A4 inhibitors (such as the antibiotic clarithromycin) cap the dose at 9 mg, and strong CYP3A4 inducers should be avoided. Give your prescriber a full medicine list.
- **Birth control pills:** the label advises a non-oral method or an added barrier method for 30 days after starting and after each dose increase. See [GLP-1s, birth control and pregnancy](/blog/glp-1-birth-control-pregnancy-2026).

## How much weight do people lose on GLP-1 pills?

Each pill has its own pivotal trials. Here is what they found.

**Wegovy tablets (OASIS 4).** 307 adults without diabetes took oral semaglutide 25 mg or placebo for 64 weeks. Average weight change was −13.6% with the tablet vs −2.4% with placebo in the label’s analysis (the NEJM paper reports −2.2% for placebo). About 3 in 4 people on the tablet lost at least 5% of their weight.

**Foundayo (ATTAIN-1 and a diabetes trial).** In ATTAIN-1, 3,127 adults without diabetes took orforglipron or placebo for 72 weeks. On the label’s tablet doses, average change was −7.4% (5.5 mg), −8.3% (9 mg) and −11.1% (17.2 mg), vs −2.1% with placebo. In a second 72-week trial in people with type 2 diabetes, the top dose gave −9.6% vs −2.5%.

The trials used capsules (6, 12 and 36 mg), while US patients get tablets. Lilly says the two aren’t substitutable mg for mg: the 36 mg capsule corresponds to the 17.2 mg tablet.

**Rybelsus (type 2 diabetes).** In the label’s 26-week monotherapy trial, HbA1c fell 1.2 points on 7 mg and 1.4 points on 14 mg, vs 0.3 with placebo. Weight fell 2.3 kg and 3.7 kg, vs 1.4 kg. In SOUL, a large heart-outcomes trial in about 9,650 people with type 2 diabetes, 14 mg lowered the risk of major heart events (hazard ratio 0.86).

### Why do news stories quote bigger numbers?

You may see 16.6% for Wegovy tablets or 12.4% for Foundayo. Those figures count people while they kept taking the drug as directed. The label numbers include everyone who started, which is closer to real life. Neither is wrong, but compare like with like.

## What are the side effects of GLP-1 pills?

The side effects mirror the injections: mostly digestive, most common while the dose is going up.

- **Rybelsus:** nausea in 11% (7 mg) and 20% (14 mg) vs 6% with placebo; diarrhea, vomiting, belly pain and lower appetite were also more common.
- **Wegovy tablets:** GI side effects in 74% vs 42% with placebo in OASIS 4, mostly mild to moderate and short-lived. Altered skin sensation (dysesthesia) occurred in 5%. 6.9% stopped for side effects vs 5.9% on placebo.
- **Foundayo 17.2 mg:** nausea 35% vs 10%, vomiting 24% vs 4%, constipation 24% vs 9% and diarrhea 25% vs 11%. 10% stopped for side effects vs 3% on placebo.

These come from different trials with different definitions, so don’t line them up as a league table. All three labels carry a boxed warning about thyroid C-cell tumors, and none of these pills is for people with a personal or family history of medullary thyroid cancer or MEN 2. For practical tips, see [how to ease GLP-1 nausea](/blog/glp-1-nausea-relief-2026).

## Should you choose a GLP-1 pill or an injection?

Neither format is better for everyone. The trade-offs your prescriber may talk through:

- **Rhythm:** a pill is daily, so there are seven chances a week to miss a dose; a shot is weekly.
- **Routine:** oral semaglutide needs a fasting morning ritual; Foundayo doesn’t; injections need a pen and site rotation.
- **Other medicines:** Foundayo has CYP3A4 limits, and oral semaglutide affects levothyroxine.
- **Evidence:** weight-loss pills have shorter track records than weekly injections. Wegovy tablets also carry a heart-risk indication; Foundayo’s label covers weight management only.
- **Combining:** the Wegovy and Foundayo labels advise against taking them with any other GLP-1 medicine.

Moving between a shot and a pill is possible, but only some labels give instructions. Our guide to [switching GLP-1 medications](/blog/switching-glp-1-medications-2026) explains what they say.

## When should you call your doctor?

Get prompt medical help if you notice:

- Severe stomach pain that doesn’t go away, with or without vomiting (possible pancreatitis)
- Vomiting or diarrhea that stops you keeping fluids down, or signs of dehydration such as dizziness and very little urine
- Pain in the upper right belly, fever or yellowing skin or eyes (possible gallbladder problems)
- Swelling of the face, lips, tongue or throat, or trouble breathing
- Shakiness, sweating or confusion if you also take insulin or a sulfonylurea (low blood sugar)
- A lump or swelling in the neck, trouble swallowing or a hoarse voice that won’t go away

## Tracking a daily GLP-1 pill

A daily pill lives or dies by routine. GLPMate logs each dose with a reminder at your chosen time, and the side-effect log with severity helps you spot patterns around dose increases. There are guides for the [Foundayo tracker](/foundayo-tracker-app) and the [Rybelsus tracker](/rybelsus-tracker-app).

FAQ

## FAQ

### Is there a GLP-1 pill for weight loss?

Yes. In the US, Wegovy tablets (oral semaglutide 25 mg, approved December 2025) and Foundayo (orforglipron, approved 1 April 2026) are approved for weight management in adults with obesity, or overweight plus a weight-related condition. Rybelsus and Ozempic tablets are oral semaglutide too, but they are approved for type 2 diabetes, not weight loss.

### Foundayo vs Wegovy pill: which works better?

There is no head-to-head trial. In separate trials, Wegovy tablets 25 mg lowered weight by 13.6% on average at 64 weeks (2.4% with placebo), and Foundayo 17.2 mg by 11.1% at 72 weeks (2.1% with placebo). The populations, durations and analyses differ, so the numbers can't be compared directly; your prescriber weighs them with your health, other medicines and daily routine.

### Do you have to take Foundayo on an empty stomach?

No. The Foundayo label says it can be taken once daily with or without food, and it sets no water limit or wait time. Oral semaglutide (Rybelsus, Ozempic tablets and Wegovy tablets) is different: it must be taken on an empty stomach with up to 4 ounces of water, then 30 minutes before eating, drinking or other oral medicines.

### Is Rybelsus the same as the Wegovy pill?

Both contain semaglutide, but they are different products with different doses and uses. Rybelsus goes up to 14 mg for type 2 diabetes, while Wegovy tablets build up to 25 mg for weight management. They aren't interchangeable, and you shouldn't take two semaglutide products together; any change between them is your prescriber's decision.
